Paul Coucke"},"content":{"rendered":"<p><strong>1. Kunt u zich alstublieft voorstellen?<\/strong><\/p>\n<p style=\"text-align: justify;\">Mijn naam is Paul Coucke. Ik ben professor aan de Universiteit Gent in het departement Medische Genetica, waar ik verantwoordelijk ben voor het bindweefsellaboratorium. Ik houd toezicht op de diagnostiek in de laboratoria, dat wil zeggen op de monsters die voor diagnostische doeleinden naar ons worden opgestuurd, en ik houd ook toezicht op het onderzoek in het laboratorium waar we onderzoek doen naar bindweefselaandoeningen, waarvan het Marfan syndroom er \u00e9\u00e9n is.<\/p>\n<p style=\"text-align: justify;\"><strong>2. Kunt u het 101 Marfan Genomes Project uitleggen?<\/strong><\/p>\n<p style=\"text-align: justify;\">Het doel van het P101GM is om fenotype-genotype correlaties vast te stellen. Dit betekent dat, nu het genoom steeds toegankelijker wordt door de ontwikkeling van sequencingtechnologie, het interessant is om te zoeken naar correlaties tussen variaties in de genen die in dit genoom gevonden worden en fenotypische expressies bij pati\u00ebnten. Dit zou verklaren waarom bepaalde pati\u00ebnten langer zijn, waarom ze een karakteristiek oogprobleem hebben, waarom ze aorta misvormingen of afwijkingen hebben. Het is erg moeilijk om verbanden te leggen tussen deze afwijkingen en de variaties in genomen. Vandaag de dag wordt het mogelijk om dit soort analyses uit te voeren, wat vijf of zelfs drie jaar geleden niet mogelijk was. Het is een grote uitdaging voor P101GM.<\/p>\n<p style=\"text-align: justify;\"><strong>3. Waarom bent u lid geworden van het wetenschappelijk comit\u00e9 van het 101 Genomes Marfan Project?<\/strong><\/p>\n<p style=\"text-align: justify;\">Ik werk nu al meer dan 30 jaar in genetisch onderzoek. Ik begon in 1985. Ik ben altijd betrokken geweest bij genetica en ik heb de evolutie ervan gezien.  <u>In de afgelopen vijf jaar is dit zeer interessant geworden door, zoals ik al zei, de ontwikkeling van genomische sequentiebepaling. Het is een technologische revolutie en de kosten voor sequentiebepaling zijn aanzienlijk gedaald, waardoor we toegang hebben gekregen tot genomische gegevens.<\/u>. De bedoeling van P101GM is om dit genoom te gebruiken in correlatie met de fenotypische kenmerken van Marfan pati\u00ebnten. Ik geloof erg in deze aanpak en ik vind dit project erg spannend.<\/p>\n<p style=\"text-align: justify;\"><strong>4. Wat verwacht je als wetenschapper van het Marfan 101 Genomes Project?<\/strong><\/p>\n<p style=\"text-align: justify;\">Het is erg moeilijk om te weten wat de uitkomst van dit onderzoek zal zijn, maar het sterke punt is dat het wetenschappers samenbrengt. De meeste wetenschappers werken in hun eigen afdelingen en laboratoria in goede faciliteiten, maar ieder in zijn eigen hoekje. Communicatie tussen afdelingen of universiteiten vindt alleen plaats tijdens vergaderingen. In werkelijkheid moeten we samenwerken, bespreken hoe we verder moeten gaan en pati\u00ebnten en pati\u00ebntgegevens samenbrengen. Maar dit is niet zo gebruikelijk in onderzoekscentra, en het zou meer moeten zijn. Dit is het voordeel van P101GM: de oprichters proberen zoveel mogelijk wetenschappers samen te brengen. Er is een competente groep uit Antwerpen, een andere zeer gespecialiseerde groep uit Gent en ook een zeer erkende groep wetenschappers uit Parijs. Afgezien van de P101GM hebben we contact met elkaar, maar dat is slechts contact. Hier hebben we de middelen om idee\u00ebn uit te wisselen, te discussi\u00ebren, samen te werken en verder te werken aan de verbetering van onderzoek.<\/p>\n<p style=\"text-align: justify;\"><strong>5. Wat is voor u het belangrijkste element dat het Marfan 101 Genomes Project belangrijk maakt voor Marfan-pati\u00ebnten? En voor andere zeldzame ziekten?<\/strong><\/p>\n<p style=\"text-align: justify;\"><u>De technische evolutie van sequencing<\/u>, die het mogelijk maakt om dingen te doen die voorheen nooit mogelijk waren. Dat is de sleutel.
